Start at Esc, not backquote
The top-left key on a 65% is Esc, because there is no function row above it. The backquote and tilde characters move onto the Fn layer. A cap that lights as Esc when you expected tilde is correct behaviour.

Work the alphanumeric block
Number row, then the three letter rows, then the bottom row. Press each key deliberately — the layout is dense and mis-hits are easy.

Right-hand column and arrows
Work down the navigation column, then press all four arrows. The up arrow sits directly beside a shortened right Shift and is the most commonly mis-hit key on the layout.

What survives on a 65% board
Sixty-five per cent keeps the alphanumeric block intact, keeps all four arrow keys, and keeps a handful of navigation keys in a column on the right — typically Delete, Page Up, Page Down and Home, though the exact set varies between manufacturers more than on any other layout.
What it drops is the entire function row. Esc moves up to where the tilde key sits on a full-size board, and the backtick and tilde characters move onto the Fn layer alongside F1 through F12.
The result is 67 testable keys and a board around 305 mm wide — roughly two-thirds of a full-size keyboard, with the arrow keys still under your right hand where you expect them.

Living without a function row
The honest version: you will notice. Anything that binds F-keys — debugging, renaming files, browser developer tools, most games' quick-slots — now needs two hands or a remap.
Most 65% owners settle into one of two habits. Either they learn the Fn combinations properly, which takes about a week and then becomes invisible, or they remap the keys they actually use onto Caps Lock and the right-hand modifiers, which is what firmware like QMK and VIA exist for.
What you get back is desk space and travel weight. A 65% fits in a laptop sleeve, which is why it has become the default choice for people who carry a keyboard between home and an office.

Where the Esc key went
On a full-size board, the top-left key of the alphanumeric block is backtick and tilde, with Esc sitting above it in the function row. Remove the function row and something has to occupy the top-left corner — so on a 65% that key becomes Esc, and backtick moves to the Fn layer.
This trips people up when a key seems dead. If you press what you think is the tilde key and Esc lights up on screen, nothing is broken; you are pressing Esc, because that is what that position now is.
Some boards ship with Esc and backtick swapped, or make the choice configurable in firmware. If the on-screen board disagrees with yours, that is why.

Testing a 65% properly
Start at the top-left with Esc and work along the number row, then the three letter rows, then the bottom row. Because the layout is dense, press each key deliberately and check which one lights up rather than assuming.
Then work down the right-hand column and finish with the four arrows. The up arrow is the one to watch: on most 65% boards it sits directly beside a shortened right Shift, and it is the single most commonly mis-hit key on the layout.
The Fn key stays greyed out throughout. That is correct — it is handled inside the keyboard and never reaches the browser, so it is excluded from the totals and a working board still reaches 100%.
